Overview

Glutathione in its reduced form (GSH) is a tripeptide — γ-L-glutamyl-L-cysteinylglycine — with the molecular formula C10H17N3O6S, a molecular weight of approximately 307.33 g/mol and CAS number 70-18-8. It is the most abundant low-molecular-weight thiol in most cell types. Its structure is unusual for a peptide: the glutamate residue is linked to cysteine through its side-chain γ-carboxyl group rather than through an α-peptide bond, which is why cells build it with two dedicated enzymes (γ-glutamylcysteine synthetase and glutathione synthetase) rather than on the ribosome, and why it resists most aminopeptidases.

The cysteine thiol is the reactive center of the molecule. In the published biochemistry GSH functions as an intracellular redox buffer, cycling between the reduced monomer and glutathione disulfide (GSSG), and serves as the reducing cosubstrate for glutathione peroxidases and the conjugating nucleophile for glutathione S-transferases. GSSG is a separate compound (C20H32N6O12S2, 612.6 g/mol) formed when two GSH molecules oxidize and join through a disulfide bond; because the free thiol oxidizes on exposure to air and moisture, the material should be kept sealed and refrigerated at 2–8 °C.
