Structurally it is an acylhydrazone: naphthalene-2-carbaldehyde condensed with 4-hydroxybenzohydrazide, giving (E)-4-hydroxy-N′-(naphthalen-2-ylmethylene)benzohydrazide. The compound existed in commercial screening libraries for years before it was identified and characterized by the Burris laboratory at Saint Louis University, from which it takes its name.

In the primary literature (Billon et al., ACS Chemical Biology, 2023) SLU-PP-332 is reported as a pan-agonist of the three estrogen-related receptors ERRα, ERRβ and ERRγ — orphan nuclear receptors involved in mitochondrial gene expression — with the highest reported potency at ERRα. The reported findings come from cultured cell lines and rodent skeletal muscle. Two handling notes follow from the chemistry: acylhydrazones can hydrolyze in aqueous acid and photoisomerize, so the powder should be kept frozen and protected from light, and the flat aromatic scaffold makes it poorly soluble in water, with DMSO the reported solvent. It is supplied as a solid powder in 5mg and 10mg vials.
